Multi-Component Switch

The intein-mediated split-Cas9 system is a multi-component CRISPR-Cas9 architecture in which Cas9 is divided into separate parts and reconstituted through split inteins. It was developed to bypass packaging limits while retaining nuclease activity comparable to wild-type Cas9, and a split-Cas9D10A version showed activity similar to Cas9D10A nickase.
